TLR4High expressionIncreased levels of TNF-α, IL-1β, IL-6, CXCL2, CXCL5, CCL5, CXCL8 in LPS-treated cells. This showed that TLR4, MyD88 and NF-κB p65 higher expression led to mammary gland infection.BMECs(Jiang et al., 2022)
Figure 1.

Bacterial-induced inflammatory signaling pathway leading to mastitis in dairy cattle. Bacterial pathogen-associated molecular patterns activated TLR4/TLR2-MyD88 signaling, which diverged into IRAK1/4-TRAF6-IKKγ/IKKα-NF-κB and MAPKK-MAPK (ERK1/2, p38, JNK) cascades. These pathways converged on nuclear transcription factors (ATF2, c-FOS/c-JUN, Elk-1, NF-κB) that induced proinflammatory cytokine expression, culminating in mammary gland inflammation. PAMP (pathogen-associated molecular pattern).

Figure 2.

Molecular pathways of stress-induced inflammatory response in mammary epithelial cells. Exogenous stressors activated TLRs and NADPH oxidase, initiating MyD88-dependent JNK/ERK/p38-MAPK signaling. ROS generation served as a central hub linking oxidative stress to IKKα/IKKβ-mediated NF-κB and AP-1 activation. Nuclear translocation of these transcription factors upregulated proinflammatory mediators, establishing a mechanistic connection between oxidative stress and mammary gland inflammation.
